Toxicity and cell density monitoring in monolayer and three-dimensional cultures with the XTT assay

The application of viability criteria (MTT and XTT tests) to monolayer cultures and immobilised cells in three-dimensional systems was investigated in order to assess cell viability and cell proliferation. The suitability and accuracy of these tests were compared with the conventional criteria (cellular protein and DNA content) used in monolayer cultures for the same purpose. The colorimetric assay based on the metabolic reduction of the tetrazolium salt XTT to a water-soluble formazan proved to be very useful, rapid and sensitive. A specific microassay for evaluating hepatic LDH activity in co-cultures of hepatocytes with other cells.

This study describes the development of a simple, rapid and reproducible microassay for determining the intracellular LDH activity of rat hepatocytes present in a co-culture system with other cells. The procedure involves treatment of cellular homogenates with an anti-LDH antiserum that specifically inhibits the LDH activity of rat hepatocytes. The assay is performed in 96-well plates and LDH activity can be measured directly in the same wells using a colorimetric method. The difference in LDH activity values measured before and after antiserum incubation reflects the LDH content of the hepatocytes in the sample. Abstract The pyridine antihistamines carbinoxamine maleate and doxylamine succinate were derivatized by hydrolysis with cyanogen bromide and coupling with aniline to form a polymethine dye, and determined by colorimetry. In a micellar cationic medium of N-cetylpyridinium chloride (NCPC), the reaction times were reduced and the sensitivities improved. In 0.1 M NCPC the apparent molar absorptivities of the derivatives were 5,900 and 3,500 M−1 cm−1, and the limits of detection (as 3s) were 8.6x10−7 M and 1.7x106 M, for carbinoxamine and doxylamine, respectively. Electrochemical methods can offer new sustainable routes for the abatement of organic pollutants resistant to biological processes. These methods use a clean reagent, the electron, and very mild operative conditions (ambient temperature and atmospheric pressure) with limited operative costs. In 2013, quinoxyfen was included in the list of priority hazard pollutants of the European Water Framework Directive due to its toxicity to aquatic organisms. However, few analytical methods for the analysis of this fungicide have been reported and no rapid immunochemical methods have been published so far. Background: The aim of this study was to evaluate high voltage electrical discharges (HVED) as a green technology, in order to establish the effectiveness of phenolic extraction from olive leaves against conventional extraction (CE).
